Synthesis And Testing Of Analogs Of Helminthosporium Maydis Race T Toxin, George A. Kraus, Michael Silveira, S.J. Danko

Several analogues of Helminthosponum maydis race T toxin were prepared. The synthesis of the hydroxy diketones was accomplished by aldol condensations of 2-decanone with ketal aldehydes followed by deketalization. The dihydroxy ketone analogues were prepared by use of dihydroisoxazole intermediates. The analogues were tested and found to be less active than the parent toxin. The analogues still exhibited host specificity.

Naphthalene-2,3-Dialdehyde: A Synthon To 7,9-Polymethylene-8h-Cyclohepta[B]Naphthalene-8-One And A Study Of Their Aromaticity, Edward O. Crapps

This report describes the synthesis of naphthalene-2,3-dialdehyde, and an analytical method for determining its purity by thin layer chromatography. Subsequently, the dialdehyde was condensed with various cyclic ketones to generate a series of polymethylene bridged naphthotropones. It was found that planarity and aromaticity increase with the size of the methylene bridge, although, the bridge must contain six or more methylene groups for the latter to be true. The claim for aromaticity is supported by NMR and IR data, which indicates the positive charge for a perchlorate cation salt is delocalized over the naphthalene and tropone ring system. A method for non-matrix corrected organic sulfur analysis by energy dispersive X-ray spectroscopy has been developed using petroleum coke standards. This technique capitalizes on the fact that when a substance is excited by an electron beam it will emit photons of electromagnetic radiation in the X-ray range. The emitted X-rays are characteristic of the excited elements in the substance. Typically, electron beam microanalysis is a rapid, nondestructive analytical technique to quantitatively measure organic sulfur in coal. In Chapter I of this dissertation, the Skattebol-type rearrangement (in THF solution at -78(DEGREES)) of anti-7-bromo-syn-7-lithiobicyclo4.1.0hept-2-ene (34-anti) and its C('7)-epimer (34-syn) is investigated. The following discoveries demonstrate that, in solution, this rearrangement proceeds through a carbenoid: (a) carbenoid 34-anti undergoes 1,3-rearrangement and generates the carbenoid coupling products 42-syn and 42-anti 17 times faster than does 34-syn, (b) the coupling products 42-syn and 42-anti are formed in a much different ratio from 34-anti (7.5 to 1) than from 34-syn (1.5 to 1), (c) no carbene trapping products were formed in the presence of ...

Several heterocyclic ring systems have been prepared by two new organometallic methods. The first approach involved the mercuration of alkyl aryl acetylenes. This reaction resulted in the formation of mercurated heterocycles. Mercurated benzofurans, isocoumarins, chromones, benzothiophenes and benzopyrans were all prepared by this method. The mercury moiety in these compounds was readily substituted by various functional groups. By making a slight modification in the acetylenic substrate, the intramolecular cyclization was prevented. The flash vacuum pyrolysis chemistry of a series of deuterated furfuryl benzoates, furfuryl-(alpha), (alpha)-d(,2) benzoate, furfuryl-5-d(,1) benzoate, and furfuryl-3-d(,1) benzoate have been investigated. Mechanisms were proposed for the formation of the pyrolysis products, methylenecyclobutenone, vinylacetylene, and the dimer of cyclopentadienone, from each of the compounds studied which involved initial alpha elimination of benzoic acid, a single 3,3 migration followed by alpha elimination of benzoic acid, and two 3,3 migrations followed by alpha elimination of benzoic acid. The Regiochemistry Of Alkenylsilyl, Alkenyldisilanyl And Alkenylsilyloxy Radical Cyclizations , Anthony Revis

The regiochemistry of intramolecular cyclization of some alkenylsilyl and alkenyldisilanyl radicals has been examined. It has been demonstrated that the silyl radicals produced by hydrogen abstraction from a pentenylsilane, a butenylsilane, a butenyldisilane, an allyldisilane, and a butenyloxysilane all cyclize in an endo-fashion. This is in complete contrast to what is known for the analogous carbon-centered radicals. No cyclic products are observed for chain lengths shorter than five atoms. Also, when alkenyldisiloxanyl and butadienyl-type silyl radicals are generated by pyrolytic cleavage of silicon-allyl bonds, endo-closure is found to be the exclusive mode of cyclization. Unpaired electron spin distribution in polysemiquinones of varying geometric disposition is discussed. Particular emphasis is placed on the dependence of spin distribution on the acceptor strength, temperature, and reduction state of the radical ions. ESR spectra of diradical species in which separation between the unpaired electrons does not exceed 10 (ANGSTROM), and which nevertheless exhibit sharp ESR transitions in non-viscous media are displayed. During the last 10 years, adriamycin and daunomycin have been major components in the treatment of cancers. However, their toxicity has led to the search for less toxic anthracycline antibiotics. The 11-deoxyanthracyclines were discovered and have proven to be more effective anticancer drugs;A program was undertaken to develop a route to the linear tetracyclic ring system of the anthracyclines. Flash vacuum pyrolysis (FVP) has been used successfully to discover some novel thermal reactions. These provide new insights into some fundamental chemistry;In part I, benzocyclobutene (9) and its 4+4 dimer, 1,5-dibenzocyclooctadiene (14) were found to give anthracene (7) as a major product. The similarity of the product mixtures is taken as evidence that essentially the same mechanism is operating in both cases. By labeling studies, the 4+2 spirodimer (14') appears to be an intermediate. The fragmentation by which 14' is thought to give 7 is new and analogous to the retroene reaction. Co-pyrolysis of 4-trimethylsilyl-1-methoxy-1-methyl-1-silacyclohexa-2,5-diene with MeOH afforded 1-methyl-1-methoxy-1-silacyclohexa-2,4-diene. Co-pyrolysis with acetylene afforded 1-methyl-1-silabicyclo2.2.2-octa-2,5,7-triene. Flash vacuum pyrolysis (FVP) gave a 4 + 2 Diels-Alder dimer of 1-methyl-1-silabenzene. These results are explained by a 1,3-trimethyl silyl shift followed by (beta)-elimination of Me(,3)SiOMe to form 1-methyl-1-silabenzene. Alkylmercury halides (RHgX, R = n-butyl, n-hexyl, (DELTA)('3)-butenyl, (DELTA)('5)-hexenyl, isopropyl, cyclohexyl, cyclopentylcarbinyl and 7-norbornyl) rapidly react with PhYYPh (Y = S, Se and Te), p-MeC(,6)H(,4)SO(,2)SePh, PhSO(,2)Cl, PhSH and BrCCl(,3) under sunlamp irradiation to give RYPh, RCl, RH and RBr, respectively. The reaction is completely inhibited by di-tert-butyl nitroxide, does not occur in the dark and yields a mixture of cyclopentylcarbinyl and (DELTA)('5)-hexenyl products from the (DELTA)('5)-hexenylmercurial.
