Many experiments have shown that the diffusive motion of lipids and membrane proteins are slower on the cell surface than those in artificial lipid bilayers or blebs. One hypothesis that may partially explain this mystery is the effect of the cytoskeleton structures on the protein dynamics. A model proposed by Kusumi [1] is the Fence-Picket Model which describes the cell membrane as a set of compartment regions, each ~ 10 to 200 nm in size, created by direct or indirect interaction of lipids and proteins with actin filaments just below the membrane. Introduction: The utility of engineered nanomaterials’ is growing, particularly the titanium dioxide (TiO2) polymorphs. TiO2 is very useful for brightening paints, and coloring foods. Nano-sized TiO2 is also useful for sunscreens, cosmetics, and can be utilized as a photocatalyst. However, the nanometer size and the large specific surface area of the TiO2 materials are physicochemical characteristics which may contribute to human red blood cell (RBC) damage. Using RBCs as a cellular model, we have evaluated the effects of TiO2 nanoparticle exposure to RBCs by quantifying oxidized glutathione, oxidized membrane vitamin E, hemolysis, hemoglobin adsorption, and cellular aggregation. Increasing global temperatures during the 21st century may have detrimental effects on basic cell functions within ectothermal animals. This project aims to systematically examine the effects of climate change on secretory cargo localization through the analysis of Rab Proteins found in the model organism Drosophila melanogaster. Rab Proteins play an essential role in vesicular transport within the cell and can be genetically manipulated to monitor the biological consequences of global warming.
