Amylotriose and amylotetraose have been separated from salivary amylase digests by carbon chromatography. Crystalline 1-phenyl flavazole derivatives of both compounds have been prepared;Comparative salivary amylase hydrolysis rates for amylotriose, amylotetraose, amyloheptaose and amylodextrin were determined;Linear radioactive oligosaccharides have been prepared by the coupling reaction of macerans amylase on alpha-dextrin and radioglucose. These have been shown to be labeled only at the reducing glucose unit. The reaction of salivary amylase on the members G3 to G7 has been studied by paper chromatography. The alpha-amylase of takadiastase can be markedly concentrated by repeated precipitation with 0.75 saturated ammonium sulfate and adsorption of precipitates by bentonite. The highest activity obtained was 145 U/mg N;Ammonium sulfate at 0.70 saturation was shown to be sufficient precipitate the alpha-amylase of takadiastase;Adsorption of alpha-amylase and limit dextrinase by rice starch and bentonite proceed in a roughly equivalent manner. An amyloclastic enzyme solution and a saccharogenic enzyme solution has been prepared from wheat extract by a method similar to that used by Ohlsson for malt extract;A solid amyloclastic enzyme with low sugar-forming power has been prepared by heating wheat extract for fifteen minutes at 70° and fractionally precipitating with alcohol;A solid saccharogenic enzyme has been prepared from wheat extract by bringing the hydrogen ion concentration to a pH of 3.3 and then adjusting it to a pH of 6.0, and finally precipitating with alcohol.
