Role Of Carbon Dioxide In The Synthesis Of Amino Acids By A Photosynthetic Bacterium In The Dark , Daniel Hartz Hug

The photosynthetic bacterium, Rhodospirillium rubrum, requires carbon dioxide for aerobic growth in the dark in a medium containing acetate. Resting cells do not oxidize acetate in the dark in the absence CO 2. Dark oxidation of acetate is stimulated by catalytic amounts of malate;Rhodospirillium rubrum when grown in a medium containing malate in the presence of radioactive carbon dioxide yields the following radioactive amino acids: aspartate, glutamate, arginine, tryptophan, proline, threonine, glycine, leucines, tyrosine, serine, methionine, valine, cystine, and lysine; hydroxyproline is tentatively identified. The literature concerning the occurrence of D-amino acids in nature, the resolution of underivatized amino acids, the resolution of glutamic acid, and the resolution of 2-aminobutan-1-ol has been reviewed;The resolution of 2-aminobutan-1-ol with (+)tartaric acid was studied;The resolution of DL-glutamic acid with (-)2-amino-butan-1-ol has been shown to be simple and reproducible;Solubility experiments showed that the diastereomers (-)2-aminobutan-1-ol-D-glutamate and (-)2-aminobutan-1-ol-L-glutamate were about equally soluble in aqueous ethanol individually. The two diastereomers together were much more soluble than would be expected from their individual solubilities. The solubilizing effect of (-)2-aminobutan-1-ol-L-glutamate on the (-)2-aminobutan-1-ol-D-glutamate was greater than the solubilizing effect ...

Preliminary experiments were performed to investigate the possibility of combining chemical blocking with microbiological assays in a method for determining terminal amino acids in peptides;A subtractive chromatographic procedure for determination of terminal amino acid residues in peptides is presented. Results are given for a total of nine dipeptides and one tripeptide. Whole cells and cell-free extracts of Aerobacter aerogenes possess an enzyme system which actively ammoniates alpha-ketoglutarate The enzyme was obtained in cell-free solution; its action was similar to that in resting cells or growing cells;The concentration of glutamate formed by growing cells in the aerobic dissimilation of a carbon source in higher than that formed under anaerobic conditions.
