A microarray was constructed and applied to transcriptional profiling and genetic variation studies of Mycoplasma hyopneumoniae. The genome sequence enabled the construction of the microarray to allow a global approach to understanding fundamental processes in M. hyopneumoniae . These studies focused on whether M. hyopneumoniae regulates its genes under different environmental conditions and if genetic changes can be correlated with virulence. Equine infectious anemia virus (EIAV) is a member of the lentivirus subfamily of retroviruses that produces a variable clinical disease course characterized as acute, chronic, and inapparent. The clinical signs can vary according to the stage of disease, and generally correlate with levels of virus replication. As with other retroviruses, EIAV utilizes both RNA and proteins to produce alternatively spliced transcripts required for virus replication. Lentiviruses are single-stranded RNA viruses generally associated with chronic diseases of the immune and central nervous systems. In contrast to the insidious, progressive nature of most lentiviral diseases, equine infectious anemia virus (EIAV) infection results in rapid onset of a variable disease course in equids. Acute disease is accompanied with high-titered viremia, thrombocytopenia, fever, depression, and inappetance. The chronic stage is usually characterized by recurrent episodes of disease. We investigated the mechanism underlying somatostatin-induced increase in [Ca2+]i leading to insulin release in the presence of arginine vasopressin (AVP) in clonal beta-cell HIT-T15. Somatostatin increased [Ca2+]i and insulin release in a biphasic pattern, characterized by a sharp and transient increase followed by a rapid decline to the sub-basal level. Pretreatment with pertussis toxin, which inactivates Gi/Go, abolished the effects of somatostatin. U-73122, an inhibitor of phospholipase C, antagonized somatostatin-induced increase in [Ca2+] i. The phospholipase C (PLPC) gene from Clostridium hemolyticum was cloned using the polymerase chain reaction. An open reading frame which encodes a 399-amino acid protein, containing a 27-amino acid signal sequence, was identified. The molecular weight of the active protein was 42,869 daltons. A 16-amino acid N-terminal sequence determined by Edman degradation exactly matched the putative amino acid sequence of the gene product. Comparison of the nucleotide and amino acid sequences with Genebank databases demonstrated that the beta toxin of C. hemolyticum exhibits high homology with other bacterial PLPCs. Genetic variation has been observed in many viruses. Viruses that carry their genetic information in the form of RNA exhibit high mutation rates because the viral polymerase lacks proof-reading mechanisms commonly found in DNA polymerase complexes. The combination of high mutation rates, small genome size, and high replication rates results in a population of closely related viral genotypes, which are commonly referred to as a quasispecies. The purpose of this study was to identify alpha-adrenoreceptor types and subtypes mediating contractile responses to adrenergic agonists in the bovine tail artery and vein and to investigate nerve-mediated contraction in the bovine tail artery in response to electrical field stimulation. Mammary Ca2+ transport and storage may contribute to the pathological hypocalcemia seen in dairy cows at parturition. Angiogenesis, the formation of new capillaries from pre-existing vessels, is essential for tumor progression. It is critical for the growth of primary cancers. In this thesis we present a new approach to angiogenesis, based on the theory of reinforced random walks, coupled with a Michaelis-Menten type mechanism. This views the endothelial cell receptors as the catalyst for transforming angiogenic factor into proteolytic enzyme in order to model the first stage. Methylamine osmolytes are enriched in urea-rich cells. It is unclear, however, whether methylamines can promote microtubule assembly either in vitro or in cells. The osmolyte trimethylamine N-oxide (TMAO) at 200 mM increases the extent and the rate of formation of polymerized microtubule complex caused by microtubule-associated protein tau in vitro. Furthermore, TMAO counteracts the inhibition on the assembly caused by urea. Atypical of other lentiviruses, such as human immunodeficiency virus type 1, equine infectious anemia virus (EIAV) may produce a rapid, variable disease course in horses. Tractin and LeechCAM are two recently identified members of the Ig-superfamily. Tractin has a highly unusual domain organization: 6 Ig-like domains, 4 FNIII-like domains, an acidic domain, 12 repeats of a novel proline- and glycine- rich motif with sequence similarity to collagen, a transmembrane domain, and an intracellular tail with an ankyrin and a PDZ-domain binding motif. ICAM-1 expression in cattle with Bovine Leukocyte Adhesion Deficiency (BLAD) vs. cattle without BLAD in Mannheimia haemolytica (MH) pneumonia at 2 or 4 hours PI was assessed using in situ hybridization. There was a significant increase in ICAM-1 in bronchiolar and alveolar epithelium and endothelium of arteries and veins in both BLAD and non-BLAD MH-inoculated cattle compared to controls. The POU protein Oct-2 has been hypothesized to partially regulate immunoglobulin gene expression through its interaction with the octamer motif found in the promoter of immunoglobulin genes. Adherence of M. hyopneumoniae to the cilia of the tracheal epithelial cells is required to establish infection. Previous studies have identified a 97-kDa protein (P97) as the putative ciliary adhesin. Mycoplasmas are deficient in numerous biosynthetic pathways. Consequently, growth is dependent upon the acquisition of structural and macromolecular precursors from the environment. These precursors include phospholipids, fatty acids and nucleotide bases. DNases and RNases are postulated to be involved in the acquisition of nucleotides. Brucella abortus strain RB51, a naturally derived rough variant, has been proposed to replace strain 19 for use as a vaccine against brucellosis in cattle. Utilizing fingerprinting and standard biotyping techniques, it was possible to differentiate RB51 from other brucellae and to ascertain the stability of RB51. A novel insulin-like immunoreactivity, adrenergic receptors (ARs) and voltage-dependent Ca[superscript]2+ channels (VDCCs) were characterized in a clonal [beta]-cell line, RINm5F, in a passage- and morphology-dependent manner. The cells exhibited two morphological features; the differentiated cells (DCs) with long neuron-like processes and the undifferentiated cells (UCs) without the processes;As morphology of the cells changed from DCs to UCs, amount of secreted insulin gradually decreased; however, insulin reacted to antiserum vs. bovine insulin (ABI) gradually increased from barely detectable amounts.
