Science, Physiology, And Nutrition For The Nonscientist, Judi S. Morrill Jul 2019

Science, Physiology, And Nutrition For The Nonscientist, Judi S. Morrill

Open Educational Resources

A wonderful blend of physiology, nutrition, biochemistry, genetics, biology, evolution, chemistry--what we all need to know as informed citizens. A basic knowledge of the life sciences and how our bodies work--to promote our own good health, especially as we're bombarded with misleading advertisements, soundbites, and the like. DNA fingerprinting, calorie requirements, dietary advice, genetic engineering (including gene editing with CRISPR cas9)--all in an easy-to understand book.

Using Fluorescence Microscopy To Identify A Potential New Treatment For Heart Failure, Ryan D. Himes Apr 2017

Using Fluorescence Microscopy To Identify A Potential New Treatment For Heart Failure, Ryan D. Himes

Scholar Week 2016 - present

Cardiac glycosides have been used to treat heart failure for centuries, but they have a narrow therapeutic window, as they inhibit their target receptor nearly irreversibly. Overdoses can lead to arrhythmias. Phospholemman is a natural inhibitor of the same target as cardiac glycosides. It is possible that mutating phospholemman could achieve the same therapeutic benefit, while allowing cells to reverse the inhibition and thereby avoid an arrhythmia. I used fluorescence microscopy to screen candidate phospholemman mutants and identify one that binds more avidly to its target than the naturally occurring phospholemman. This mutant, L30A, caused similar effects as cardiac glycosides ...

Expression Of Bovine Mammary Gland Ca2+Atpases , Siriwan Prapong Jan 2000

Expression Of Bovine Mammary Gland Ca2+Atpases , Siriwan Prapong

Retrospective Theses and Dissertations

Control of transcellular Ca2+ fluxes associated with lactation are critical to mammary function and Ca2+ homeostasis. Mammary Ca2+ transport and storage may contribute to the pathological hypocalcemia seen in dairy cows at parturition. RT-PCR and Western Blotting were used to identify which Ca2+ ATPases are present in the bovine mammary gland and in milk fat globule membranes (MFGM). PMCA1, PMCA2 and PMCA4 are plasma membrane Ca2+ ATPases expressed in the bovine mammary gland. A putative Golgi Ca2+ ATPase (bSPCA) is also found in significant amounts in the mammary gland. The expression of Ca2+ ATPases in the bovine mammary gland is ...

The Role(S) Of Arginine-Specific Mono(Adp-Ribosyl)Transferase In Skeletal Muscle Cells , Hui-Yu Huang Jan 1995

The Role(S) Of Arginine-Specific Mono(Adp-Ribosyl)Transferase In Skeletal Muscle Cells , Hui-Yu Huang

Retrospective Theses and Dissertations

The overall goal of this research project was to examine the biological function of arginine-specific mono(ADP-ribosyl) transferase in skeletal muscle cells. To better understand the function of this enzyme in skeletal muscle, the experimental approach to accomplish this goal consisted of two specific objectives. The first objective was to identify the protein substrates for this enzyme in myogenic cell cultures and cellular fractions. Embryonic chick primary muscle cell cultures were used as a working system for this purpose. In membrane fractions of 96-h myotube cultures incubated with [[superscript]32 P]NAD under conditions selected for mono(ADP-ribosyl)ation, proteins ...

The 5Å Resolution Crystal Structure Of Adenylosuccinate Synthetase From Escherichia Coli , Michael Alan Serra Jan 1990

The 5Å Resolution Crystal Structure Of Adenylosuccinate Synthetase From Escherichia Coli , Michael Alan Serra

Retrospective Theses and Dissertations

The technique of X-ray crystallography has been employed to determine the structure of adenylosuccinate synthetase from Escherichia coli to a resolution of 5A. Three crystalline forms of the enzyme have been obtained. The structure determination of the P21 crystal form was pursued due to its excellent diffraction properties and its rapid and reproducible growth. The P21 form crystallizes as a dimer. Unit cell parameters are as follows: a = 73.31(9)A, b = 72.23(7)A, c = 82.87(9)A, [beta] = 108.59°(6). The unit cell volume is 416,700(1,200)A[superscript]3.;A suitable ...
