Retinoic Acid Glucuronide Preparations For Application To The Skin, Arun B. Barua, Desiree Gunning, James A. Olson

A method and preparation are described for treating human skin with retinoic acid glucuronide (RAG) in a topical carrier. For treatment of acne or wrinkled skin, RAG can be applied in an effective amount which is nonirritating to the skin. Retinoid dermatitis, an objectionable side effect of topical application of retinoid compounds, can thereby be avoided.

For treatment of acne the glucuronide of retinoic acid (retinoyl β-glucuronide) is orally administered to women of child-bearing age who are capable of conception. This therapy is safer than the oral administration of retinoic acid because retinoic acid glucuronide is essentially non-teratogenic at dose levels effective for the acne treatment. The retinoic acid glucuronide can be in all-trans or 13-cis form.

The cDNA clones encoding for the bovine glycoproteins CD44 and LECAM-1 (lectin cell adhesion molecule-1) were isolated and sequenced. The predicted amino acid sequences of bovine CD44 and LECAM-1 had high identities with those of human and murine CD44 and LECAM-1. The high identities of the predicted amino acid sequences of these glycoproteins in the bovine, human and mouse suggest they may have similar functions in all three species. Using encoding schemes, we study the relation between amino acids and protein structure in linear spaces;Local protein-structure prediction schemes use the amino acid sequences in a short subchain of a protein to predict the protein secondary structure of the middle residue of the chain;Two relatively reliable, objective and quantitative local prediction schemes are Robson et al.'s information theory method and and Qian & Sejnowski's neural network models. Partial physical maps of each of the three replicons of Rhizobium meliloti were constructed using restriction enzymes PacI, SpeI, and AseI. Using transverse alternating field gel electrophoresis (TAFE) in combination with very long pulse times, low agarose concentrations, and low field gradients, or in combination with PacI restriction enzyme digestions, the three replicons of R. meliloti 1021 were separated. Three bands were separated from undigested genomic DNA samples using TAFE; Southern hybridizations with representative cloned genes from each replicon revealed that each band corresponded to one of the replicons. The purpose of this research is to determine the role that post-transcriptional processes play in the regulation of phytochrome (phy) steady-state messenger RNA (mRNA) levels. The control of etiolated phytochrome (phyA) and chlorophyll a/b binding protein (cab) gene transcription and steady-state mRNA levels are known to be regulated by the phyA signal transduction pathway. The effect of the chelator 1,10-phenanthroline on the DNA-binding of 1,25-dihydroxyvitamin D receptor was demonstrated. Increasing concentrations of the chelator, up to 3 mM, decreased receptor binding to DNA-cellulose. This effect was specific for the chelating function because the nonchelating analog, 1,7-phenanthroline, did not inhibit DNA-cellulose binding of the receptor. The cations, Cd[superscript]2+ and Zn[superscript]2+, were able to block chelator-mediated inhibition. The regulation of genes by CO[subscript]2 concentration in wild type and mutants of the unicellular green alga Chlamydomonas reinhardtii was studied. This and other algal species can photosynthesize efficiently under limiting concentrations of CO[subscript]2 due to the activities of a CO[subscript]2 concentrating mechanism and carbonic anhydrase. A potato Proteinase Inhibitor II gene (pin2T) was isolated and characterized both at the molecular level and the functional level. The open reading frame as well as the 5[superscript]' and 3[superscript]' flanking regions were sequenced. The coding region and 3[superscript]' flanking region showed 86% and 87% identity with those of the previously isolated wound-inducible potato Proteinase Inhibitor IIK gene (pin2K) sequence, respectively. However, the 5[superscript]' flanking region of the pin2T is highly homologous (91% identity) with that of pin2K from -767 to +29, relative to the transcription start site of the wound-inducible pin2K.
