Photodynamic Therapy Of Murine Fibrosarcoma With Topical And Systemic Administration Of Hypericin, I Cavarga, P Brezani, Maria Cekanova, P Solar, P Fedorocko, P Miskovsky

Maria Cekanova MS, RNDr, PhD

The in vivo antitumour activity of the natural photosensitizer hypericin was evaluated. C3H/DiSn mice inoculated with fibrosarcoma G5:1:13 cells were intraperitoneally or intratumourally injected with hypericin (5 mg/kg) and 2 hours later the mice were locally irradiated with laser light (488 nm, 150 mW/cm2, 180 J/cm2) when the tumour reached volume of 40-80 mm3 (approximately 17 days after inoculation). Tumours treated with hypericin alone as well as those irradiated with laser light alone have similar growth rates and none of these tumours regressed spontaneously. The mean tumour volume in hypericin-PDT treated groups was significantly ...

Mathematical Modeling Of The Onset Of Capillary Formation Initiating Angiogenesis, Howard Levine, Brian D. Sleeman, Marit Nilsen-Hamilton Mar 2001

Mathematical Modeling Of The Onset Of Capillary Formation Initiating Angiogenesis, Howard Levine, Brian D. Sleeman, Marit Nilsen-Hamilton

Mathematics Publications

It is well accepted that neo-vascular formation can be divided into three main stages (which may be overlapping): (1) changes within the existing vessel, (2) formation of a new channel, (3) maturation of the new vessel.

In this paper we present a new approach to angiogenesis, based on the theory of reinforced random walks, coupled with a Michaelis-Menten type mechanism which views the endothelial cell receptors as the catalyst for transforming angiogenic factor into proteolytic enzyme in order to model the first stage. In this model, a single layer of endothelial cells is separated by a vascular wall from an ...

Investigation Of The Effect Of Bcl-2 On Hybridoma Cell Survival , Mark Christopher Mowry Jan 2001

Investigation Of The Effect Of Bcl-2 On Hybridoma Cell Survival , Mark Christopher Mowry

Retrospective Theses and Dissertations

Hybridoma cells are widely used to produce large quantities of monoclonal antibodies for use as medicines and diagnostics. Bcl-2 is an oncogene that has been shown to have an anti-apoptotic effect in many cell lines including hybridomas. This study investigates the effect of various levels of bcl-2 expression on hybridoma cell grown and antibody productivity in both batch and perfusion culture.;H166 hybridomas were transfected with a plasmid containing a bcl-2 insert and stable clones were selected. The cells were grown in spinner flasks in batch mode for twelve days and it was confirmed that expression at high levels allow ...

Coordination Of Cell Morphology And Nuclear Division By The Bud Neck Control Ring In Saccharomyces Cerevisiae , Courtney Lynn Thomas Jan 2001

Coordination Of Cell Morphology And Nuclear Division By The Bud Neck Control Ring In Saccharomyces Cerevisiae , Courtney Lynn Thomas

Retrospective Theses and Dissertations

Coordination of daughter cell growth and nuclear division are readily observed but not well understood in the yeast Saccharomyces cerevisiae . A screen was performed to identify mutants which lost this coordination such that they exhibited an elongated cell morphology (improper cell growth), but continued to divide. The results of this screen and suppressor analysis suggests the bud neck ring is a control structure which determines cell morphology as well as nuclear division as an upstream regulator of Cdc28p-cyclin complexes.;The effects of mutations in specific components of the bud neck ring on the assembly of other components into the structure ...

Drosophila Jil-1 Kinase Mediates Histone H3 Ser10 Phosphorylation, Maintains Higher Order Chromatin Structure, And Is Implicated In Dosage Compensation , Yanming Wang Jan 2001

Drosophila Jil-1 Kinase Mediates Histone H3 Ser10 Phosphorylation, Maintains Higher Order Chromatin Structure, And Is Implicated In Dosage Compensation , Yanming Wang

Retrospective Theses and Dissertations

The Drosophila JIL-1 tandem Ser/Thr kinase is associated with chromosomes/chromatin throughout the cell cycle in early embryos and localized to hundreds of sites on the open interband regions on third instar larvae polytene chromosomes. Interestingly, the level of JIL-1 is upregulated on the male X chromosome, which is hypertranscribed for dosage compensation. The distribution of JIL-1 overlaps with that of MSL (male specific lethal) proteins and JIL-1 is associated with the MSL complex.;To further study the function of JIL-1 in vivo, a series of JIL-1 mutants from hypomorphs to null were generated. Analyzing the phenotypes of JIL-1 ...

Characterization Of The Biological Activities Of Recombinant Fusion Protein Green Fluorescent Protein/Human Zona Pellucida Protein 3 (Gfp/Hzp3), Zhiyong Lin Jan 2001

Characterization Of The Biological Activities Of Recombinant Fusion Protein Green Fluorescent Protein/Human Zona Pellucida Protein 3 (Gfp/Hzp3), Zhiyong Lin

Theses and Dissertations in Biomedical Sciences

Despite numerous reports indicating the successful production of bioactive recombinant ZP3, no report has shown the rhZP3 having direct binding activity with human sperm. Recombinant ZP3 generated from our previous study displayed binding activity with human sperm through indirect evidence from hemizona assay (HZA).

This present study focused on the production of recombinant ZP3 with direct binding activity with human sperm. Through the application of a pEGFP expression vector, fusion protein GFP/ZP3 was successfully generated and expressed. The expression of GFP/ZP3 was evidenced by RT-PCR and western blot. The fusion protein was partially purified by Ni-NTA affinity column ...

The Paradox Of Plastid Transit Peptides: Conservation Of Function Despite Divergence In Primary Structure, Barry Bruce Dec 2000

The Paradox Of Plastid Transit Peptides: Conservation Of Function Despite Divergence In Primary Structure, Barry Bruce

Barry D. Bruce

Transit peptides are N-terminal extensions that facilitate the targeting and translocation of cytosolically synthesized precursors into plastids via a post-translational mechanism. With the complete Arabidopsis genome in hand, it is now evident that transit peptides direct more than 3500 different proteins into the plastid during the life of a typical plant. Deciphering a common mechanism for how this multitude of targeting sequences function has been hampered by the realization that at a primary sequence level, transit peptides are highly divergent in length, composition, and organization. This review addresses recent findings on several of the diverse functions that transit peptides must ...

Abnormal Expression Of P27kip1 Protein In Levator Ani Muscle Of Aging Women With Pelvic Floor Disorders - A Relationship To The Cellular Differentiation And Degeneration., A Bukovsky, P Copas, M Caudle, Maria Cekanova, T Dassanayake, B Asbury, S Vanmeter, R Elder, J Brown, S Cross

Maria Cekanova MS, RNDr, PhD

BACKGROUND: Pelvic floor disorders affect almost 50% of aging women. An important role in the pelvic floor support belongs to the levator ani muscle. The p27/kip1 (p27) protein, multifunctional cyclin-dependent kinase inhibitor, shows changing expression in differentiating skeletal muscle cells during development, and relatively high levels of p27 RNA were detected in the normal human skeletal muscles. METHODS: Biopsy samples of levator ani muscle were obtained from 22 symptomatic patients with stress urinary incontinence, pelvic organ prolapse, and overlaps (age range 38-74), and nine asymptomatic women (age 31-49). Cryostat sections were investigated for p27 protein expression and type I ...
