Phosphatase of Regenerating Liver (PRL) proteins regulate a number of important cellular processes, including cell growth and division. Humans have three PRL proteins: PRL-1, PRL-2, and PRL-3. An accumulation of evidence has shown that elevated levels of PRLs are strongly correlated with uncontrollable growth and metastasis of tumors. However, contradictory findings have arisen indicating that PRLs instead function to halt cell division thereby preventing uncontrollable tumor growth. In light of these results, the underlying mechanisms regarding how PRLs function within cellular processes remains unclear. ABSTRACT: The HIV-1 pandemic continues to thrive due to ineffective HIV-1 vaccines. Historically, the world’s most infectious diseases, such as polio and smallpox, have been eradicated or have come close to eradication due to the advent of effective vaccines. Highly active antiretroviral therapy is able to delay the onset of AIDS but can neither rid the body of HIV-1 proviral DNA nor prevent further transmission. p53 is a transcription factor that plays a significant role in cell cycle regulation, senescence, and apoptosis. The p53 protein has been widely studied for its link to human cancer, and is estimated to be mutated in ~50% of all human cancers. This project focused on generating a p53-null A549 cell line using the genome editing technique CRISPR (Clustered Regularly-Interspaced Short Palindromic Repeats)-Cas9. CRISPR-Cas9 was employed to generate a simultaneous knock-in/knockout of mCherry and WTp53, respectively.
