Enhancing genetic resistance of chickens to Newcastle Disease Virus (NDV) provides a promising way to improve poultry health, and to alleviate poverty and food insecurity in developing countries. In this study, two inbred chicken lines with different responses to NDV, Fayoumi and Leghorn, were challenged with LaSota NDV strain at 21 days of age. Through transcriptome analysis, gene expression in spleen at 2 and 6 days post-inoculation was compared between NDV-infected and control groups, as well as between chicken lines. Fast-growing modern broilers are susceptible to cardiac arrhythmia and heart failure. To understand the developmental regulation related to these problems, cardiac gene expression in Illinois and Ross broilers was investigated at 6 and 21 days post hatch (dph) using RNA-seq (n=5-6 for each group). Because Illinois is a legacy broiler line with isometric heart growth, and Ross is a modern broiler line with a negative allometric heart growth, comparison of gene expression between the two lines and the two time points provides an avenue to understand genetic regulation underlying their different cardiac development.

Modern fast-growing broilers are susceptible to heart failure under heat stress because their relatively small hearts cannot meet increased need of blood pumping. Velogenic Newcastle disease virus (NDV) causes global economic losses in the poultry industry. Although proper vaccination does prevent outbreaks of ND, genetic selection for enhanced resistance to NDV offers a complementary approach to disease prevention. Understanding the gene expression changes induced by ND infection will help to elucidate mechanisms of host resistance and susceptibility.

Newcastle disease virus (NDV) is an infectious virus in domestic chickens and is endemic in many countries. It has caused huge economic losses worldwide, especially in developing countries where the chickens and eggs are a primary source of economic income and family nutrition for the rural poor. The objective of this study was to investigate the effect of mild heat stress on muscle fiber hyperplastic and hypertrophic growth in quail primary myogenic cells to better understand the mechanisms leading to increased skeletal muscle development in avian embryos incubated at a higher temperature. Compared to control cultures maintained at 37°C, incubation at 39°C enhanced myotube length (P < 0.01) and diameter (P < 0.001) at 3 days after differentiation (D3). The fast growth rate and increased body weight of modern broilers makes them susceptible to heart failure and sensitive to heat stress. To find the genetic causes of these problems, gene expression was measured in hearts of two broiler lines that differed in their response to heat stress. Prolonged exposure to high temperature can significantly increase metabolism, depress appetite, reduce meat production and raise mortality in broilers. Because the cardiovascular system plays an important role to dissipate heat and transport oxygen, strong cardiac function is indispensable for birds to survive under heat stress. In this study, CCNG2 and CDKN2C were found to be correlated with cellcycle inhibition during fat cell differentiation, whereas CCND3, CCNA1, and ANAPC5 were positively associated with cellcycle activity during fat cell proliferation after selection based on GEO datasets available on the NCBI website. Three single nucleotide polymorphisms (SNPs) at -660bp (deletion of a single guanine base), -380bp (T>C), -69bp (C>T) were identified by sequencing cDNA of CYB5 genes in different Chinese and Foreign pigs. After subsequent detection using microsatellite, pyrosequencing and restriction fragment length polymorphism (RFLP) techniques in large populations of different pig breeds, we found that the mutations in all the SNP sites were favorable in Chinese pigs except LiCha black pigs which were crossed with Landrace pigs before, whereas the wild type nucleotides were favorable in Foreign pigs including Large White, Landrace and Duroc pigs.
