West Nile virus (WNV) infection results in a spectrum of neurological symptoms, ranging from a benign fever to severe WNV neuroinvasive disease with high mortality. Many who recover from WNV neuroinvasive infection present with long-term deficits, including weakness, fatigue, and cognitive problems. While neurons are a main target of WNV, other cell types, especially astrocytes, play an important role in promoting WNV-mediated central nervous system (CNS) damage. Background: Predicting prognosis in patients from large-scale genomic data is a fundamentally challenging problem in genomic medicine. However, the prognosis still remains poor in many diseases. The poor prognosis maybe caused by high complexity of biological systems, where multiple biological components and their hierarchical relationships are involved. Moreover, it is challenging to develop robust computational solutions with high-dimension, low-sample size data. Background: Reconstruction of gene regulatory networks (GRNs), also known as reverse engineering of GRNs, aims to infer the potential regulation relationships between genes. With the development of biotechnology, such as gene chip microarray and RNA-sequencing, the high-throughput data generated provide us with more opportunities to infer the gene-gene interaction relationships using gene expression data and hence understand the underlying mechanism of biological processes. The U. S. Gulf of Mexico is experiencing a dramatic increase in tidal marsh restoration actions, which involves planting coastal areas with smooth cordgrass (Spartina alterniflora) and black needlerush (Juncus roemerianus) for erosion control and to provide habitat for fish and wildlife. Bacterial infections and antibiotic resistance, particularly by Gram-negative pathogens, have become a global healthcare crisis. We report the design of a class of cationic antimicrobial polymers that cluster local facial amphiphilicity from repeating units to enhance interactions with bacterial membranes without requiring a globally conformational arrangement associated with highly unfavorable entropic loss. This concept of macromolecular architectures is demonstrated with a series of multicyclic natural product-based cationic polymers. Background

The health and resilience of species in natural environments is increasingly challenged by complex anthropogenic stressor combinations including climate change, habitat encroachment, and chemical contamination. To better understand impacts of these stressors we examined the individual- and combined-stressor impacts of malaria infection, food limitation, and 2,4,6-trinitrotoluene (TNT) exposures on gene expression in livers of Western fence lizards (WFL, Sceloporus occidentalis) using custom WFL transcriptome-based microarrays.

Advancements in wearable technology have facilitated performance monitoring in a number of sports. Figure skating may also benefit from this technology, but the inherent movements present some unique challenges. The purpose of this study was to evaluate the feasibility of using an inertial measurement unit (IMU) to monitor three aspects of figure skating jumping performance: jump count, jump height, and rotation speed. Carboxypeptidase O (CPO) is a member of the M14 family of metallocarboxypeptidases with a preference for the cleavage of C-terminal acidic amino acids. CPO is largely expressed in the small intestine, although it has been detected in other tissues such as the brain and ovaries. The microbiome plays an essential role in the health and onset of diseases in all animals, including humans. The microbiome has emerged as a central theme in environmental toxicology, as microbes interact with the host immune system in addition to its role in chemical detoxification. Critical habitat was designated in 2003 for federally threatened anadromous Gulf sturgeon to aid in population recovery. This study examined overwintering Gulf sturgeon spatial use and movement through critical habitat monitored by the Ship Island acoustic array from 2011 to 2015. Previous studies observed western population Gulf sturgeon (Pearl and Pascagoula rivers) overwintering near the ends and within the passes of the barrier islands of the Mississippi Sound, USA. Background: Mesenchymal stem cells (MSCs) isolated from adult tissues (Ad-MSCs) have shown great promise for use in regenerative medicine. However, their poor in vitro expansion capacity and tissue scarcity have been major limitations. In this study, we demonstrate that mouse embryonic stem cells (mESCs) can differentiate into cells with MSC properties.

Methods: Using previously established methods that characterize Ad-MSCs, we analyzed mESC-differentiated fibroblasts (mESC-FBs), including plastic adherence, clonogenic growth, MSC marker expression, tri-lineage differentiation potential, and the capacity to express immunomodulators.

Even though others exist, CrossFit training has emerged as the most common form of HIFT. Background: We investigated interactions between genetic and psychological factors in predicting shoulder impairment phenotypes. We hypothesized that pro-inflammatory genes would display stronger relationships compared with pain-related genes when combined with psychological factors for predicting phenotypic changes.
Subjects and methods: Altogether, 190 participants completed a 5-day experimental protocol. An experimental shoulder injury model was used to induce physical impairment, and a priori selected genetic (pain-related, pro-inflammatory) and psychological (anxiety, depressive symptoms, pain catastrophizing, fear of pain, kinesiophobia) factors were included as predictors of interest. The main objective of this study was to test comparatively the effects of two common insecticides on honey bee Apis mellifera worker’s lifespan, food consumption, mortality, and expression of antioxidant genes. Newly emerged worker bees were exposed to organophosphate insecticide coumaphos, a neonicotinoid imidacloprid, and their mixtures. Toxicity tests were conducted along with bee midgut immunohistological TUNEL analyses. RT-qPCR assessed the regulation of 10 bee antioxidant genes linked to pesticide toxicity. This study investigated the effect of branched-chain amino acid (BCAA) supplementationon recovery from eccentric exercise. Twenty males ingested either a BCAA supplement or placebo(PLCB) prior to and following eccentric exercise. Creatine kinase (CK), vertical jump (VJ), maximalvoluntary isometric contraction (MVIC), jump squat (JS) and perceived soreness were assessed.No significant (p> 0.05) group by time interaction effects were observed for CK, soreness, MVIC, VJ,or JS. CK concentrations were elevated above baseline (p< 0.001) in both groups at 4, 24, 48 and 72 hr,while CK was lower (p= 0.02) in the BCAA group at 48 hr compared to PLCB. Soreness increasedsignificantly from baseline (p< 0.01) in both groups at all time-points; however, BCAA supplementedindividuals reported less soreness (p< 0.01) at the 48 and 72 hr time-points. An inability to lift loads great enough to disrupt muscular blood flow may impair the ability to fatigue muscles, compromising the hypertrophic response. It is unknown what level of blood flow restriction (BFR) pressure, if any, is necessary to reach failure at very low-loads [i.e., 15% one-repetition maximum (1RM)]. The Atlantic surfclam (Spisula solidissima) is a dominant member of the biological community of the Middle Atlantic Bight continental shelf and a commercially harvested species. Climate warming is affecting the biology and distribution of this species, which provides an opportunity to investigate the processes and conditions that are restructuring this fishery and the implications for ecological and socioeconomic systems. Inflammation is known to alter nervous system function, but its effect on muscle spindle afferent mechanosensation and sensory integration in the spinal cord has not been well studied. We tested the hypothesis that systemic inflammation induced by an intraperitoneal injection of the endotoxin lipopolysaccharide (LPS; 7.5 × 105 endotoxin units/kg 18 h before experiment) would alter muscle spindle afferent mechanosensation and spinal cord excitability to Group Ia input in male and female adult C57Bl/6 mice. Sharks and rays are popular species used in wildlife ecotourism and aquariums to educate the public on the behavior, ecology and conservation challenges of elasmobranchs. Resistance training may differentially affect morphological adaptations along the length of uni-articular and bi-articular muscles. The purpose of this study was to compare changes in muscle morphology along the length of the rectus femoris (RF) and vastus lateralis (VL) in response to resistance training. Pathogen colonization inside tick tissues is a significant aspect of the overall competence of a vector. Amblyomma maculatum is a competent vector of the spotted fever group rickettsiae, Rickettsia parkeri. When R. parkeri colonizes its tick host, it has the opportunity to dynamically interact with not just its host but with the endosymbionts living within it, and this enables it to modulate the tick’s defenses by regulating tick gene expression. A small percentage of babies born to Zika virus (ZIKV)-infected mothers manifest severe defects at birth, including microcephaly. Among those who appeared healthy at birth, there are increasing reports of postnatal growth or developmental defects. However, the impact of congenital ZIKV infection in postnatal development is poorly understood. Here, we report that a mild congenital ZIKV-infection in pups born to immunocompetent pregnant mice did not display apparent defects at birth, but manifested postnatal growth impediments and neurobehavioral deficits, which include reduced locomotor and cognitive deficits that persisted into adulthood. Background: Shipwrecks serve as a rich source for novel microbial populations that have largely remained undiscovered. Low temperatures, lack of sunlight, and the availability of substrates derived from the shipwreck’s hull and cargo may provide an environment in which microbes can develop unique metabolic adaptations.

Background: Specific mood states were examined during the CrossFit Open, a consecutive 5-week fitness competition involving five separate CrossFit®workouts, to determine whether the unique design or strenuous workouts of the competition resulted in acute and/or chronic mood statealterations. To determine the salivary steroid response to high-intensity functional training (HIFT)competition workouts, saliva samples were collected from ten recreationally trained male and femalecompetitors during a 5-week (WK1–WK5) international competition. Competitors arrived at theirlocal affiliate and provided samples prior to (PRE) their warm-up, immediately (IP), 30-min (30P),and 60-min (60P) post-exercise. Samples were analyzed for concentrations of testosterone (T), cortisol(C), and their ratio (TC). Generalized linear mixed models with repeated measures revealed significantmain effects for time (p< 0.001) for T, C, and TC. Compared to PRE-concentrations, elevated (p< 0.05)T was observed at IP on WK2–WK5 (mean difference: 135–511 pg·mL−1), at 30P on WK3 (meandifference: 81.0±30.1 pg·mL−1) and WK5 (mean difference: 56.6±22.7 pg·mL−1), and at 60P onWK3 (mean difference: 73.5±29.7 pg·mL−1) and WK5 (mean difference: 74.3±28.4 pg·mL−1).Compared to PRE-concentrations, elevated (p< 0.05) C was noted on all weeks at IP (mean difference:9.3–15.9 ng·mL−1) and 30P (mean difference: 6.0–19.9 ng·mL−1); significant (p< 0.006) elevationswere noted at 60P on WK1 (mean difference: 9.1±3.0 ng·mL−1) and WK5 (mean difference:12.8±2.9 ng·mL−1). Additionally, TC was significantly reduced from PRE-values by 61% on WK1 at60P (p= 0.040) and by 80% on WK5 at 30P (p= 0.023). Differences in T, C, and TC were also observedbetween weeks at specific time points. Although each workout affected concentrations in T, C, and/orthe TC ratio, changes appeared to be modulated by the presence of overload and workout duration.During periods of elevated training or competition, athletes and coaches may consider monitoringthese hormones for consistency and as a means of assessing workout difficulty.
