Nationally, Maine is the state with the second highest incidence of Lyme disease. While the spread of Lyme disease is generally attributable to ecological factors that affect the life cycle of Lyme-spreading ticks, socioeconomic factors may have substantial impacts on diagnosis and reporting of human cases. The dystroglycan protein is one of many that attach skeletal muscle fibers to the basement membrane at the myotendinous junction. In the alpha-subunit of the dystroglycan molecule, there are sugar chains that help with the adhesion of the molecule to the basement membrane. A mutation in any gene that codes for an enzyme that adds these sugar chains can result in a form of congenital muscle disease called secondary dystroglycanopathy. Streptococcus agalactiae, otherwise known as Group B Streptococcus (GBS), is a zoonotic, Gram-positive, commensal and invasive bacteria which is the leading cause of neonatal bacterial infections. These bacterial infections include sepsis, meningitis, pneumonia, and bacteremia. In neonates, GBS is most commonly transferred to the child in utero or during birth when the child aspirates amniotic or vaginal fluids. GBS can also infect the child through the bloodstream while in utero, causing premature births or still births. The black-legged tick (Ixodes scapularis) has recently made a tremendous impact in Maine due to its role as a vector for the bacterial pathogen Borrelia burgdorferi, the causative agent of Lyme disease. A lesser known, but equally concerning, invasive insect is the hemlock woolly adelgid (HWA; Adelges tsugae), a sap-sucking scale that is primarily responsible for the ongoing widespread decline of eastern hemlock in the northeast. Forest harvesting can impact the environment in many ways, one of which is causing a loss of subsidies and increased light intensity to freshwater ecosystems. This can have a major impact on freshwater invertebrate communities that may rely on subsidies to survive. In this study, I tested two effects of commercial clearcut, changes in light availability and detrital resources, on freshwater invertebrate communities. Unsexual Salamanders within the Blue-Spotted Salamander Complex carry combinations of ambystomatid genomes (those of Blue-Spotted Salamanders, Ambystoma laterale, and Jefferson Salamanders, A. jeffersonianum in Maine). They are nearly all female, breed in wetlands, and use sperm of related species to reproduce. Little is known about their ecology to guide the conservation of this unique lineage. I examined breeding site occupancy, demographics, orientation, and terrestrial habitat selection of Unisexual Salamanders in comparison to Blue-Spotted Salamanders and other amphibians. Most marine organisms are broadcast spawners, releasing their sperm and eggs into the water column. Methods of measuring in situ fertilization have proven successful with a few model species, which are reviewed in my introductory chapter. However, many commercially exploited species, such as the sea scallop Placopecten magellanicus, have been neglected. Ruffed grouse (Bonasa umbellus) occupy a wide distribution in North America, from Georgia in the south to Alaska in the north, as well as southern and central Canada. Color phases in ruffed grouse range from red to gray with gray phase birds found more frequently at higher latitudes than red birds. This intervention would provide evidence, to pre-medical students, that social science training is important during medical school. Semi-structured interviews were conducted of six doctors from hospitals in the Maine. The State of Maine issues an unlimited number of commercial permits for the harvest of White Suckers Catostomus commersonii in Maine’s inland waters. The fishery provides a necessary source of fresh lobster Homarus americanus bait to coastal communities at a time when other bait sources are scarce. The impacts of the increasing number of permits and subsequent numbers of fishermen on the white sucker population is unknown. Muscle cells must anchor to their environment, the extracellular matrix (ECM), in order to function properly. Muscular dystrophy occurs when muscle cells cannot anchor to the ECM because specific protein complexes, such as the dystroglycan complex, are disrupted. Previously published studies have shown that overexpressing Paxillin can reduce dystrophy in dag1 deficient embryos. The aim of this study is to determine which domains of Paxillin are necessary to rescue dystrophy by overexpressing Paxillin with the LD2 or LD4 domains knocked out in dystrophic embryos. Chapter 1 of this manuscript is a literature review that serves as an introduction to the entire dissertation. Chapter 2 examines the effects of the melatonin injection on heart rate and rhythmicity in Drosophila melanogaster Canton-S (wild-type) pupae and pupae bearing a variety of heart mutations. Chapter 3 investigates further the possible mechanisms of melatonin’s ability to increase heart rhythmicity without significantly affecting heart rate. A melatonin antagonist, luzindole; a high-affinity melatonin agonist, 2-iodomelatonin and RNAi techniques are used to identify a possible melatonin receptor in Drosophila melanogaster.

Conservation of forest-dependent amphibians is dependent on finding a balance between timber management and species’ habitat requirements. To examine the effect of short-term vegetative regrowth post-harvesting on amphibian habitat use, we studied the response of eight species (four forest specialists and four habitat generalists) to four forestry treatments (partial harvest, clearcut with coarse woody debris [CWD] removed, clearcut with CWD retained, and uncut control) over a 6-year period, using replicated experimental treatments in Maine, USA. Conservation of forest amphibians is dependent on finding the right balance between management for timber production and meeting species' habitat requirements. For many pond-breeding amphibians, successful dispersal of the juvenile stage is essential for long-term population persistence. We investigated the influence of timber-harvesting practices on the movements of juvenile wood frogs (Lithobates sylvaticus). We used a chronosequence of stands produced by clear-cutting to evaluate how stand age affects habitat permeability to movements. In this dissertation I investigate the impacts of both growth management regulations and vernal pool protection regulations on landscape permeability for wood frogs (Rana sylvatica), a vernal pool-obligate species. Motivated by new legislation in Maine (USA) to protect vernal pools and the species that depend on them, I combined a spatially-explicit economic model of the conversion of undeveloped parcels to residential use with a landscape permeability model for wood frogs. Relative abundance of amphibians in forest canopy gaps of natural origin vs. timber harvest origin.— Small–scale canopy gaps created by logging may retain adequate habitat structure to maintain amphibian abundance. We used pitfalls with drift fences to measure relative abundance of amphibians in 44 harvested gaps, 19 natural treefall gaps, and 36 closed–canopy forest plots. Metamorphs had relatively lower capture rates in large harvest gaps for Ambystoma maculatum, Lithobates catesbeianus, L. clamitans, and L. sylvaticus but we did not detect statistically significant (p < 0.1) differences among gap types for Lithobates palustris metamorphs. Compared to other phocids seals, the maternal investment strategy of the small bodied female harbor seal (Phoca vitulina) is complex. Females are unable to fast for the entire duration of pup rearing and are therefore reliant on resources in the vicinity of the pupping aggregation to continue provisioning their pup by mid-lactation. At the same time, harbor seal pups are highly active during lactation which increases energetic costs to the female but also offers an opportunity for females to influence the behavioral development of her pup. Since the early 19801s, the numbers of adult Atlantic salmon (Salmo salar) returning to Maine's rivers have been in a general decline. In addition, estimates of parr, freshwater smolt and emigrating smolt populations indicate low overwinter survival rates. Overall, these low numbers, along with several other factors, resulted in the Atlantic salmon in eight rivers in Maine being classified as a distinct population segment under the Endangered Species Act.
